Development of Pramipexole Prolonged-Release Matrix Tablets

The present work deals with the development of a prolonged release matrix tablet readily soluble drug pramipexole. new excipient Kollidon SR (a mixture 80% polyvinyl acetate and 20% polyvinylpyrrolidone) hypromellose K15M were used as polymers to control from selected prototypes. Various polymer concentrations tested achieve stable pH independent pramipexole 0.26 mg over 24 hours; samples prepared by wet granulation compression tablets. Drug was monitored in dissolution media values 1.2 6.8 compared reference product based on hydrophilic combination three different polymers. required profile low-dose formulation achieved using 60.0% (w/w) all F2 demonstrated sufficient alcohol resistance up ethanol concentration 40% (v/v). Predložená práca sa zaoberá vývojom matricovej tablety s predĺženým uvoľňovaním ľahko rozpustného liečiva pramipexolu. Ako polymér riadiaci uvoľňovanie z vybraných prototypov bol použitý nový (zmes 80 % polyvinylacetátu 20 polyvinylpyrolidónu) hypromelóza K15M. Testovali rôzne koncentrácie polymérov cieľom dosiahnúť stabilné nezávislé pramipexolu 0,26 po dobu hodín, vzorky boli pripravené vlhkou granuláciou lisovaním matricových tabliet. Uvoľňovanie bolo sledované v disolučných médiách hodnotami od 1,2 až 6,8 porovnávané referenčným prípravkom na báze hydrofilnej matrice kombináciu troch rôznych polymérov. Požadovaný liberačný profil nízkodávkového dosiahnutý použitím 60,0 hm.% Kollidonu vo všetkých vybraná formulácia vykazovala dostatočnú odolnosť aj voči pôsobeniu etanolu do 40 obj.%.
